ESTIMATION OF PLASMA/SERUM BIOCARBONATE

REAGENT

  1. NORMAL SALINE

COMPOSITION

Saline                                           1g

Distilled water                            100ml

PREPARATION

Dissolved 1g of Sodium chloride in 100ml of 100ml water

  1. 01N (HCL) HYDROCHLORIC ACID
  2. (NaOH) SODIUM HYDROXIDE 01N

COMPOSITION

Distilled water

Sodium hydroxide

 

PREPARATION

Dissolve 0.1NaOH in distilled water, make 1: 10 dilution.

 

 

  1. INDICATOR SOLUTION

COMPOSITION

Phenol red                                                        0.1g

0.5NaOH (sodium hydroxide)                         6.0ml

Distilled water                                                  100ml

 

PREPARATION

Dissolved 0.1g of Phenol red in 6.0ml 0.5NaOH (sodium hydroxide) and dilute it with 100ml of distilled water

 

METHOD

Set up two tube as showing in the table below

 

REAGENT CONTROL TUBE TEST TUBE
Serum 0.1ml 0.1ml
Normal saline 5.0ml 4.0ml
Indicator solution 2 drop 2 drop
0.01NHCL 1.0ml 1.0ml
0.01NaOH   Drop by drop till the colour change from yellow to red

STEP 1

Preparation of a control tubes as follow

Add 5.0ml of normal saline in a control tube; add 0.1ml of serum to the tube, add 2 drop of phenol red indicator mix very well.

The colour should correspond to the end point of the titration

STEP 2

Add 4.0ml of normal saline in a test tube, add 1.0ml of 0.0NHCL follow with 0.1ml of serum, add 2 drop of phenol red indicator.

Titrate against 0.0N NaOH till the colour change from yellow to red.

Then compare the colour of the test tube with the control tube. See table above for practical example.

NB: the reading is in ml si unit

CALCULATION

(1-Xml of 0.01N NaOH requird) x 100
